Force and conductivity measurements with single DNA molecules

Following an introduction to single molecule manipulation of nucleic acids, we present an experimental study of the electrical conductivity of double stranded DNA. Single DNA molecules are manipulated with sub piconewton force and deposited on gold nanoelectrodes by a dual optical trap. The DNA is modified at its ends for specific bead attachments and along the chain to favour charge transfer between the DNA base pair stack and the electrodes. For an electrode separation of 70 nm we find, in aqueous environment, electrical resistances above 100 GOhm.
